Envolvimento de receptores opióides e endocanabinóides da substância negra na modulação da atividade da via GABAérgica nigro-tectal durante a organização do comportamento de defesa eliciado por roedores confrontados com serpentes veneníferas

Resumo

Há um grande interesse científico nas bases neuropsicofarmacológicas dos comportamentos que têm sido asociados ao medo inato e ao pânico. Muitos estudos sugerem que o teto mesencefgálico (TM) modula as respostas defensivas elaboradas durante situações ameaçadoras. As camadas profundas do colículo superior (cpCS) têm sido consideradas uma importante estrutura na elaboração de tais reações comportamentais. A substantia nigra, pars reticulata (SNpr) é conhecida por enviar projeções inibitórias para as camadas intermediárias (ciCS) e profundas do colículo superiors modulando o padrão de respostas de defesa organizadas por exemplo pela substância cinzenta periaquedutal dorsal. Por outro lado, o TM envia projeções para as divisões compacta e reticulada da substância negra, por meio de vias neurais ricas em peptídeos opioides endógenos. O mesencéfalo ventral ainda possui receptores endocanabinoides do tipo CB1 e CB2, que podem contribuir para a modulação do comportamento de defesa. Portanto, o propósito do presente trabalho é estudar o papel de receptores opioides do tipo µ e kappa, assim como o de receptores endocanabinoides CB1 e CB2, da divisão reticulada da substância negra, rica em neurônios GABAérgicos de projeção nigro-tectal, na modulação do comportamento de defesa que tem sido relacionado ao medo e ao pânico. Contudo, coexistem tanto receptores opioides clássicos, como também receptores da nociceptina na SNpr, e sua participação na atividade da via nigro-tectal também merece investigação. Dessa forma, serão estudados os efeitos de microinjeções de agonistas e de antagonistas opioides seletivos para os receptores µ1 e kappa, de antagonistas endocanabinoides CB1 e CB2, e antagonistas seletivos do receptor ORL1 na SNpr sobre a elaboração do comportamento de defesa organizado pelo mesencéfalo dorsal e induzido pelo antagonismo seletivo de receptores GABAA nas cpCS de Ratos Wistar.
