Estimation and evaluation of withdrawal periods for albendazole and its metabolites in fish

Abstract

Currently, aquaculture is the fastest growing food production system in the world. The use of veterinary drugs is of great importance in any system of animal production to treat and prevent diseases. However, there are few drugs regulated in Brazil to be administered to aquaculture species. Due to the lack of alternatives for veterinary drugs and the high cost of available medicines, there is a suspicion that fish farmers are resorting to irregular use of prescribed substances for other animal species, or even banned substances. Such practice may lead to the presence of residues of these substances in food, posing a risk to consumer\'s toxicological safety. Albendazole (ABZ) is a broad-spectrum anti-helminth, which is widely used for cattle, sheep and goats, and it is already proven effective for fish, but its use is not yet approved for fish farming. Considering the potential of ABZ as a drug alternative for use in aquaculture systems and its process of regulation, the present project aimed to: (i) incorporate ABZ in feed through a suspension with 5% gelatin solution (w/w) and (ii) carry out a residual depletion study of ABZ and its main metabolites and estimate the withdraw period in tilapia and patinga fillets using high performance liquid chromatography coupled to mass spectrometry (LC-MS/MS). Suspension of ABZ with 5% (w//w) gelatin solution allowed incorporation into the feed without altering the physical structure of the same, allowing dose homogeneity and low leaching rates to water. ABZ was isolated from the feed by liquid-liquid extraction. ABZ and its metabolites were extracted from fish fillets (muscle and skin in natural proportions) by QhEChERS procedure. Chromatographic separation was performed by hybrid octadecyl reverse phase column. To evaluate reliability of the method, analytical validation was performed, based on recommendations VICH GL49 guide. The methods were evaluated by the following parameters: selectivity, analytical curves, linearity, precision and accuracy. All parameters are according to the validation guides. For ABZ depletion study, fishes received a single dose of 10 mg of ABZ (kg live weight)-1. The curve of residual depletion of ABZ and its metabolites in patinga fillets adjusted to the quadratic model, but it was not possible to construct the residual depletion curve for tilapia.
