Abstract

Industrial scale production conditions of biotechnology products of commercial interest, therapeutic and industry depends significantly on the separation and purification techniques used. Polymer aqueous two-phase systems (SPDFA) application is proposed as an alternative to purification, because it allows the separation and analysis of biomolecules/bioparticle, decreasing the lose its activity or properties. This technique is interesting for purification in large scale, it allows selective partition, with the potential to obtain high yields, and provide cost-effective. The work aims to study the concentration of virus adenovirus type Ad5 by extraction in aqueous two-phase system using a novel aqueous polymeric system, composed of the polymer polyethylene glycol (PEG) and polyacrylic acid (NAPA), which will be studied, using experimental design, polymer aqueous PEG / NaPA different compositions, employing different molecular weight of PEG (2000, 4000 and 8000 g / mol) and NaPA (1200, 8000 and 15000 g / mol) and concentrations of 10, 15 and 20% for both as well as the influence of pH (6.0, 7.0 and 8.0) and temperature (25, 35 and 45 ° C). The influence of these variables extraction in the concentration of adenovirus (type Ad5) will be evaluated and after this step, the extraction conditions will be optimized by better extraction and concentration of the virus in an only phase of the system.
