Abstract

Methylphenidate is widely used in the treatment of children and adolescents with attention deficit hyperactivity disorder. Nicotine is the main active substance of tabacco and is often used in combination with psychostimulants. Behavioral sensitization is a progressive and enduring enhancement of the motor stimulant effects of psychostimulants and other drugs of abuse, elicited by repeated administration of these drugs. This phenomenon has been extensively studied in experimental animals and has been implicated as a key component in drug addiction. The aim of the present study will be to investigate the behavioral sensitization induced by metylphenidate in rats exposed to repeated treatment with the this psychostimulant during adolescence and if the treatment with methylphenidate during adolescence will produce cross-sensitization when a challenge dose of nicotine is administrated during adolescence and adulthood. Thus, the adolescent rats day post natal P25-P35 will receive daily administration of 10.0 mg/kg of methylphenidate or saline by oral administration during 5 days, on P39 the methylphenidate or saline-induced locomotor activity will be registered for 60 minutes. On P40 and P70 the animals pretreated with methylphenidate or saline will receive 0.4 mg/kg nicotine or saline subcutaneous injections and the locomotor activity will be registered for 30 minutes.
