Abstract

Changes in the pattern of gene expression are responsible for many of the phenotypic differences found between species. An important category of regulatory molecules that have attracted researchers attention in the last few years are the non-coding RNAs. Despite the fact that they have already been identified in archaea, the ncRNAs participation in the regulatory process of the third domain of life is not well established yet. This work aims to promote the characterization of some non-coding RNAs of Halobacterium salinarum, an obligate halophilic archaeon. To reach this goal, ten ncRNAs modulated under H. salinarum growth curve will be selected and their 5' and 3' ends will be precisely determined. These ncRNAs will be overexpressed and their function will be evaluated in a global manner by comparing the mRNAs levels in the wild type and the strains overexpressing the ncRNAs of interest. In addition, the structural and the target predictions of the selected non-coding RNAs will be determined using bioinformatics tools. The resulting data will be analyzed and will contribute to refine the global gene regulatory network of Halobacterium with mechanistic information and to unravel the role of ncRNAs in the third domain of life.
