Abstract

This Scientific Initiation project aims at studying the growth of the gram positive bacterium Bacillus megaterium, expressing the gene pac, of the enzyme penicillin G acylase.Kinetic parameters of cellular growth will be estimated, using the cultivation medium of Yang, 2007, as a basis.Shaker assays will be run, followed by experiments in an automated bioreactor of 2L. PGA, produced extra-cellularly by B. megaterium, is an enzyme of great industrial importance, used mainly for the production of 6-aminopenicillanic acid, 6-APA, from the hydrolysis of microbial penicillin G. The enzyme that is most frequently used in industry is produced in the periplasmatic space of E. coli, what increases the downstream costs. This is an important competitive advantage of B. megaterium. The research group of the advisor of this Scientific Initiation, Prof. Roberto de Campos Giordano (Laboratory for Development and Automation of Bioprocesses, LaDABio), has been developing and applying for years techniques for fitting non-linear models, and, therefore, the student has access to an extensive library of in-house algorithms, that will be tested for fitting the cellular kinetic growth model. This investigation will have the close cooperation of the Laboratory of Enzymatic Processes Engineering, coordinated by Prof. Raquel Giordano, and will be co-advised by Dr. Monica Iemma, researcher of that group that is presently cloning the gene pac of B. megaterium in the B. megaterium itself.
