Abstract

Glucose, essential for cellular functionality, is regulated by catabolic hormones, such as glucagon, and anabolic hormones, such as insulin. Glycemic control also involves the autonomic nervous system, especially the paraventricular nucleus of the hypothalamus (PVN), integrating neuroendocrine and metabolic responses. Neurons from the PVN project to the dorsal motor nucleus of the vagus (DMV) which regulates liver functions. Previous studies have demonstrated that intracerebroventricular (ICV) insulin injection reduces hepatic vein glucose through activation of the ANS parasympathetic pathway. Therefore, our hypothesis is that ICV insulin microinjection activates neurons in the PVN which, in turn, stimulate vagal neurons that reach the liver using oxytocin as a neurotransmitter in this process. To test this hypothesis, we will perform in vitro neuroanatomy studies by tracing neural pathways between PVN-DVM and identifying neurons activated through FOS protein. We will also carry out the neurochemical characterization of the brain nuclei activated by ICV insulin. In in vivo studies, we will use pharmacological tools and OT receptor antagonism in the DMV to evaluate their effects on hepatic vein blood glucose concentration after ICV insulin injection.
