Abstract

Quality control in the pharmaceutical industry has a great socioeconomic impact, especially when it comes to a product generated from a bioprocess, which presents greater challenges than traditional chemical synthesis approaches to monitor, control, and optimize the manufacturing process in a way that guarantees its final quality. With this objective, it is necessary to develop analytical methodologies that allow the reliable measurement of critical parameters. In this context, fluorescence spectroscopy presents itself as a suitable technique for monitoring the production of virus-like particles (VLP) when combined with multivariate statistical tools. or artificial neural networks. VLP are promising candidates for the development of a prophylactic vaccine against the Zika virus, which presents a global threat due to the consequences of its infection, but there is still no approved treatment. The present project is dedicated to the use of fluorescence spectroscopy data in offline monitoring of VLP-Zika bioreactor production, correlating them using partial least squares (PLS) and artificial neural network (ANN) multivariate analysis techniques. In this way, models with predictive capacity will be generated to quantify critical process parameters such as nutrients (glucose, glutamine, glutamate), metabolites (lactate, ammonium), the density of viable cells, cell viability, and viral titer. The established models can be used to optimize and monitor VLP-Zika production, being able to reduce cost and analytical time when applied throughout the technological development process and on an industrial scale.
