Abstract

The evaluation of the biological behavior for the formation of mineralized tissue of reparative materials in direct contact with dental pulp is of paramount importance in the field of dentistry. These materials are commonly employed in restorative procedures, where it is crucial to protect and preserve the integrity of dental pulp. The aim of this study will be to evaluate in vivo the biological behavior and potential for mineralized tissue formation of bioceramic materials, compared to calcium hydroxide, when used in direct pulp capping procedures. Tissue reaction will be analyzed in the first upper molars of rats. Rats will be randomly divided into 4 groups: Bio-C Repair, CIMMO DTA, calcium hydroxide, and control group (CG, healthy molars without intervention). The pulp chamber of the first upper molars of the animals will be perforated with a 1/4 spherical bur; in this exposed area of the pulp tissue, one of the materials will be placed. The coronary chamber will be cleaned and restored with light-cure glass ionomer cement (VitremerTM, 3M ESPE Dental Products, Saint Paul, USA). In the healthy group, the first molars without any treatment will be used as controls. After 7, 21, and 45 days, right maxillary fragments will be fixed, decalcified, and processed for paraffin embedding. The volume density (Vv) of inflammatory cells (VvIc), fibroblasts (VvFb) and odontoblasts-like cells (VvOd) will be estimated in H&E-stained sections. Interleukin-6 (IL-6) and TNF-± (pro-inflammatory cytokines), and osteopontin will be detected by immunohistochemistry/immunofluorescence reactions. Data will be statistically analyzed using GraphPad Prism 8.4.3 software and subjected to ANOVA and Tukey tests (pd0.05).
