Abstract

Oral squamous cell carcinoma (SCC) is the most common oral cancer in the world and an estimated 370,000 new cases per year. Survival rates remain low, below 50% after 5 years of diagnosis and with up to 78% rates of recurrence. Thus, new potential therapeutic targets are urgently needed to be discovered to assist in therapeutic strategies. Recently, our group has unprecedentedly validated four splicing factors associated with lymph node metastasis in CEC. In this project, we aim to use structural proteomics strategies, such as proximity labeling and cross-linking agents to identify the interaction partners, as well as to evaluate the interaction interfaces between the proteins in the targeted cells and their respective extracellular vesicles. Synthetic peptides will be designed from the interfaces and tested in functional experiments with CD4+ lymphocytes and macrophages. This project will allow assessing the immune response associated with lymph node metastasis upon targeted protein modulation, indicating pathways or proteins that can be considered as potential therapeutic target in oral squamous cell carcinoma.
