Abstract

The bacterium Acinetobacter baumannii represents a serious threat in Intensive Care Units (ICUs), especially due to its ability to form biofilms and resist several antibiotics. Therefore, it is crucial to investigate the antibacterial potential of herbal products as an alternative treatment for these infections. In this study, the antibacterial and antibiofilm effects of barbatimão extract (Stryphnodendron adstringens) will be evaluated against four multidrug-resistant clinical strains and a standard strain of A. baumannii. For this, the extract will be prepared and the soluble solids content will also be quantified. Subsequently, the content of flavonoids and total phenols will be quantified, antioxidant analysis will be carried out through the reduction of the 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical, and phytochemistry will be carried out using liquid chromatography (HPLC). The minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) will be determined using the broth microdilution test according to the CLSI guidelines (standard M7-A9). Subsequently, the antibiofilm action of the extract will be evaluated by the MTT test, using previously established minimum bactericidal concentrations (CBM), after 5 min and 24 hours of treatment on already formed biofilms. Data analysis will be carried out using the Graphpad Prism 5.0 software, with the ANOVA test complemented by the Tukey test, considering a significance level of 5% (p<0.05).
