Abstract

This study will investigate the effect of functionalization of zirconia discs with collagen associated with hydroxyapatite using an in vitro implantology model and human osteoblasts. Zirconia discs with 8mm-diameter will be obtained and standardized by sand blasts (400, 600, 1200 and 2000). Then, discs will be sterilized in autoclave and subjected to funcionalization with collagen type I (1mg/mL), associated or not to hydroxyapatite (2,5). Polished zirconia discs will be used as control. After incubation at 37oC for 4 hours, cells will be seeded onto the different surfaces at 2x104 cells using complete DMEM supplemented with antibiotic solution and 10 of fetal bovine serum, After 7 days, cell adhesion will be evaluated by scanning electron microscopy cell viability will be determined by prestoBlue assay, while alkaline phosphatase activity will be evaluated by an end point assay. Results will be analyzed according to distribution and homocedasticity and then subjected to appropriate statistical analysis, considering 5 significance level.
