Cumulative effect of high potency bisphosphonate on alveolar bone repair of young female 5-LOKO mice

The chronic administration of bisphosphonates containing nitrogen associated to invasive procedures in oral cavity can lead to a condition named osteonecrosis of the jaws (ONJ), which is influenced by biological sex and age of the individual. The present study aims to evaluate bone repair process of post-extraction dental sockets of young female 129 Sv/Ev mice with alteration in 5LO gene (129Alox5tm1Fun/J) (5LOKO), and without the alteration (Wild Type - WT), which will be distributed in the following groups: WT and 5LOKO, treated with 0.01 ml of sterile 0.9% saline solution via intraperitoneal (IP), and WT-ZL and 5LOKO-ZL, treated with 250µg/Kg of zoledronic acid IP, diluted in sterile saline solution, both administrated once a week. After the fourth dose of both solutions, the animals will undergo the extraction of the upper right incisors, continuing to be medicated until the euthanasia that will be after 7, 21 and 45 days of the surgeries (n=5 per group and period). After the experimental periods, the animals will be euthanized for the removal of the maxillae with the sockets that will be evaluated under descriptive microscopical analysis from the slices stained with HE, as well as under histometric quantification of the cells and structural components in the healing area. Quantitative data will be treated by statistics, considering p<0.05.
