In the present project we propose conducting an unprecedented study based on rational engineering of the lipase-producing species Aspergillus niger, previously studied by our group, with a view to developing new, highly enantioselective enzymes. The genome of Aspergillus includes many genes of enzymes that have yet to be investigated and therefore potentially concealing new lipases to be used by industry. The means to be employed include bioprospection for genes and the production of recombinant lipases using the Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR/Cas9) technique, purification of new enzymes, and their posterior application in enantioselectivity models with racemic molecules. The project can be expected to expand the knowledge and selection of new enzymes with improved biochemical and kinetic properties as potential catalysts for the fine chemicals industry. (AU)
