Abstract

Dental implants with osteogenic potential loaded with Calcium (Ca) and Strontium (Sr) may be promising to modulate bone turnover towards osteogenesis and accelerate osseointegration. In this sense, the candidate's master's project, supported by FAPESP - process 2022/08006-7, is dedicated to evaluate the effect of the addition of Ca, Sr and osteogenic biomolecules - dexamethasone, ascorbic acid and beta-glycerophosphate, on commercially pure titanium surface. Physicochemical characterizations of the new surfaces are being carried out. The present project aims to perform a complementary analysis in the biological characterization of the new surfaces. The protein expression will be evaluated using MC3T3-E1 pre-osteoblastic cell line by enzyme-linked immunosorbent assays (ELISA). The results of gene expression of 84 osteogenic markers that are being evaluated by PCR Array will be used for the selection of 3 prominent markers that will be elected for protein analysis, in the periods of 3 and 7 days of cell culture. Statistical analyses will be performed in GraphPad Prism 5.0 software using parametric or nonparametric tests for independent data and more than two samples (ANOVA in case of normal sample distribution and homogeneity of variances or Kruskal-Wallis test), followed by multiple comparison tests, when applicable.
