TELOCYTES AND PERISINAPTIC SCHWANN'S CELLS: REPERCUSSION ON THE NEUROMUSCULAR JUNCTION IN AN EXPERIMENTAL MODEL OF MUSCLE INJURY

Abstract

The neuromuscular junction (NMJ) is the denomination of the region of communication between the peripheral nervous system (motor neuron), the sarcolemma of the muscle fiber, and the perisynaptic Schwann cells (PSC). Changes in the NMJ trigger adaptations in its morphology and repercussions on motor performance, in addition, they directly influence the structure of its pre-and postsynaptic regions, and possibly on its adjacent cells. The present study aims to reveal the remodeling of neuromuscular junctions and the possible influence of their adjacent cells in adult Wistar rats submitted to muscle injury, thus verifying the possible association of telocytes with perisynaptic Schwann cells. 56 adult male Wistar rats divided equally into 8 groups: Control (C), will use the contralateral limb of the animals in the Muscular Injury (MI) groups of 0h, 24h, 48h and 7 days post injury. The samples of the gastrocnemius muscle will be processed for the following techniques: Light Microscopy for analysis of damage in the muscle tissue and in the connective tissue through Hematoxylin and Eosin (HE) and Picrosirius (PS) staining; Immunofluorescence for the identification and measurement of pre (Basson) and postsynaptic (Q-bungarotoxin) regions of NMJ, perisynaptic Schwann cells (S100-²) and telocytes (PDGFR-Q/CD34+) in this region; will be used flow cytometry technique associated with the level of gene expression of the telocytes and the perisynaptic Schwann cells for molecular mechanisms analysis, acetylcholine receptors and elements associated with the synaptic signaling pathway through real-time PCR. The results may contribute by revealing in which periods adaptations occur in neuromuscular junctions, in addition to the identification of telocytes in this region and possible association with perisynaptic Schwann cells.
