Evaluation of bone repair after use of genistein functionalized bone substitutes to fill peri-implant defects: preclinical study in ovariectomized rats

Abstract

The search for the ideal bone substitute has driven great advances in the field of Biomaterials in Dentistry. With the emergence of biomaterials that satisfactorily perform their osteoconductive role, we see the need to seek a biological improvement of these products, through the addition of biomolecules that can have a positive effect on reparative events, improving their properties. The objective of this project will be to evaluate, through biomechanical, molecular, microtomographic, histomorphometric and laser confocal microscopy analysis, the performance of local genistein associated with Biogran®, Bio-Oss® and Cerabone®, in the process of bone repair in peri-implant defects, through the homogenization promoted by the sonochemistry technique, in ovariectomized rats. For this, 70 female rats will have a peri-implant defect in the tibial metaphysis, the peri-implant defect will be filled with biomaterial functionalized with genistein, except for the positive and negative control groups. The animals will be divided positive, they will be randomly distributed 7 groups (n=10). G1 will be the negative group while G2, G3 and G4 will be the positive control groups: G1 (clot group), G2 (Bio-Oss® without genistein), G3 (Cerabone® without genistein), G4 (Biogran® without genistein), G5 (Bio-Oss® and genistein), G6 (Cerabone® and genistein), G7 (Biogran® and genistein). After a period of 28 days postoperatively, the animals will be euthanized and the proposed analyzes will be performed. The data will be submitted to a normality curve to determine the appropriate statistical test in the GraphPad Prism 7.03 software, adopting the significance level P < 0.05.
