EVALUATION OF LOCAL THERAPIES FOR THE PREVENTION OF JAW OSTEONECROSIS IN OVARIECTOMIZED RATS TREATED WITH ZOLEDRONIC ACID. IN VIVO STUDY

Abstract

Osteoporosis is a systemic and multifactorial skeletal disease, characterized by increased bone fragility, which mainly affects postmenopausal women, and whose treatments include the use of bisphosphonates. The objective of this project will be to evaluate the use of ozonated oil and doxycycline as prevention of OMIM after extraction of the lower right first molar in ovariectomized rats treated with zolendronic acid. Forty ovariectomized adult Wistar rats will be used, which will be treated with 36 applications of 0.066 mg/kg of zoledronic acid (ZOL) intraperitoneally for 12 weeks, followed by the extraction of their right lower molars. Immediately after extraction, the animals will receive local therapies in the dental socket. The animals will be divided into five groups (n=8) according to the established therapies: Control Group (CG), will be simulated ovariectomy and treatment with saline solution (NaCl 0.9%); Ovariectomy and Serum Group (OVX-S), ovariectomy and treatment with saline solution (NaCl 0.9%) will be performed; Ovariectomy and Zoledronate Group (OVX-Z), ovariectomy and treatment with zoledronic acid will be performed; Ozonized Oil Group (GO), ovariectomy, treatment with zoledronic acid and local application of ozonized oil will be performed; Doxycycline Group (GD), ovariectomy will be performed, treatment with zoledronic acid and will be treated with filling of the socket with 10% doxycycline gel. All animals will receive intramuscular application of calcein 13 days and alizarin 23 days after extraction. At 28 days after tooth extraction, the animals will be euthanized for evaluation through: 1) microtomographic analysis and 2) confocal laser microscopy. The data obtained will be submitted to statistical analysis with p< 0.05.
