Abstract

CENTD has been focusing on the study of new inflammatory targets as well as on the screening for anti-inflammatory compounds from our library of peptides derived from venoms. Our group recently identified potential venom-derived peptides with anti-inflammatory activity in established cell models for screening. In order to identify the molecular targets of these peptides in cell models, a chemoproteomics platform was established in the Laboratory. Thus, the peptides were modified and synthesized with photo-leucine and biotin for an in vivo cross-linked chemoproteomic analysis by UV radiation and additional co-purification using streptavidin columns. Peptide targets have been identified using mass spectrometry. To assess the functional role of each molecular target in the anti-inflammatory context, a validation and gene editing platform was established at CENTD. Thus, the main objective of this project is to carry out the functional study and validation of the molecular targets identified in chemoproteomics through gene editing assays by CRISPR/Cas9. (AU)
