Chromatin damages can be caused by a series of endogenous and exogenous factors that lead to alterations and breaks on DNA, replication forks collapse, histones and other nucleus' proteins damage, besides other organelles damages such asmitochondria's with consequent increase of oxygen and nitrogen reactive species (ORSs and NRSs). Failure in appropriate cellular response during genotoxic stress can lead to genomic instability with consequent loss of cellular viability. Previous studies showed that during genotoxic stress on Saccharomyces cerevisiae, multiples proteins present an increasing level and alter their cellular localisation. It was observed that Yhb1 (ortholog of human neuroglobin NGB), a flavohemoglobin with nitric oxide reductase activity, has its levels increased during genotoxic stress induced by the genotoxic agent MMS. Besides, previous data using quantitative proteomic approach shows that after MMS treatment, there was an enrichment of Yhb1 in mitochondrial fraction. Therefore, we intend to investigate the role of Yhb1 during genotoxic stress and a possible cross-talk between mitochondrial function.
