Evaluation of the osteoconductive potential of BoneCermic® and Plenum-Oss® biomaterials in critical defects in rat caps: histometric, immunohistochemical and microtomographic analysis.

Abstract

Loss of bone tissue is often found in the dental clinic, being a factor that limits rehabilitation with implants. In this sense, synthetic biomaterials such as Plenum-oss® have been the subject of some studies and seem to show promising results in bone repair. The objective of this work is to evaluate the osteoconductive potential of Plenum-oss® and BoneCeramic®, in critical defects in the cap of rats. For that, 27 male Wistar rats, between 450 and 500g will be submitted to bilateral critical defect in the parietal region. The animals will be divided into three groups, with 9 animals (18 defects) per group: group 1 blood clot (negative control), group 2 BoneCeramic® (positive control) and group 3 Plenum-Oss®. 3 animals from each group will be euthanized after 7 days of grafting, for histometric analysis of newly formed bone, connective tissue and remaining biomaterial in the region of the critical defect and the immunohistochemical parameter of VEGF (Vascular endothelial growth factor). At 14 days, 3 more animals from each group will be euthanized to evaluate the same histometric and immunohistochemical parameters for VEGF and OC (Osteocalcin). At 28 days, the other 3 animals from each group will be euthanized to perform micro-tomography for the parameters of BV/TV, Po.(tot), Tb.N, Tb.Sp and Tb.Th, in addition histometric and immunohistochemical (OC) parameters will be evaluated. The data obtained will be submitted to a normality test to establish the best statistical analysis, considering p<0.05.
