The great importance of understanding the interaction of Hsp90 relies on the fact that this chaperone is a 'hub protein' that interacts with at least 10% of the proteins expressed in eukaryotes. Hsp90 interacts with the R2TP system and the Hsp70/Hsp40 system, all of which are involved in genomic and protein homeostasis (proteostasis). Tah1 and Pih1 connect the RVBs, which have helicase function, with Hsp90. Hsp40 binds and delivers unfolded proteins to the Hsp70/Hsp90 system. However, little information exists about the interaction regions due, among other less preponderant factors, to the high molecular mass of Hsp90, the fact that some interactions are transient and the difficulty of obtaining some partner proteins in high concentration for studies. We want to map and understand the details of interactions between Hsp90 and other proteins (chaperones, co-chaperones and clients). For this purpose we will combine protein engineering, hydrodynamics and small angle light scattering (SAXS) techniques to generate conformational models of the interaction. Furthermore, we will investigate the effect that post-translational modifications may have on the activity and interactions of these proteins, also investing in the production of recombinant proteins using insect cells.
