Influence of different premature ovarian failure models on dental socket repair of C57Bl/6J female mice treated with high potency bisphosphonate

Abstract

Premature ovarian failure (POF) causes a number of physiological changes that importantly manifest in bone tissue, leading to the development of osteoporosis. Usually, osteoporosis treatment is based on antiresorptive drugs, as the contain nitrogen bisphosphonates (nBPs), that although they are efficient for the treatment of osteolytic conditions, they are also related to the development of osteonecrosis of the jaws, especially when associated to risk factors as the oral surgeries. However, there are different possibilities of inducing experimental POF using animal models. The aim of the present study is to evaluate the impact of surgically and chemically induced POF associated to the administration of zoledronic acid (ZL) on dental socket repair after tooth extraction. Thirty-six female C57Bl/6J mice will be used, distributed into 6 groups according to the treatment: Control - no treatment; OVX - ovariectomized (OVX); VCD - treated with 160mg/Kg de VCD (4-vinylcyclohexene diepoxide); ZL-treated with 50µg/Kg of ZL; OVX+ZL - OVX treated with ZL; VCD+ZL - Treated with VCD and ZL. All the animals will undergo the extraction of the upper incisor tooth and be euthanized after 21 days. The sockets will be analyzed qualitatively and quantitatively under optical microscopy in the histological slices stained with HE, and also prepared for immunohistochemistry technique for RUNX-2 and TRAP. Quantitative data will be analyzed under statistical tests, considering p<0.05.(AU)
