Abstract

RNA is a macromolecule involved in the processes of cell transcription and translation. In recent years, it has been shown that non-coding RNAs (ncRNAs) can act in several mechanisms of cell regulation, from gene expression to chromosomal rearrangements. Thus, a class of 200-1000 nucleotids ncRNAs, the so-called long non-coding RNAs (lncRNAs) has been drawing the attention of the scientific community due to their participation in cellular mechanisms, such as embryonic development and maintenance of stem cell in various mammalian tissues. In addition, lncRNAs are abundant and some are exclusively expressed in the testis, which indicates its role in spermatogenesis. The current project aims to identify lncRNAs in spermatogonia and somatic cells of zebrafish testis through RNA sequencing, and further evaluation of their possible role in spermatogenesis, focusing on spermatogonial stem cell self-renewal and differentiation through knockout and knockdown techniques. Finally, this project will evaluate the possible role of lncRNAs as biomarkers of environmental contamination. In this sense, this project aims to provide information about the importance of lncRNAs in regulating the spermatogenic process in fish and the biological effects that certain environmental contaminants can bring to the reproductive health of such animals.
