Abstract

Patient-derived xenografts (PDX) are a relevant approach to the study of glioblastomas (GBM) since these cells mimic tumor characteristics in vitro. GBM is the most aggressive and difficult-to-treat primary brain tumor, and its high malignancy and resistance to therapy are due, among other factors, to a subpopulation of tumor stem cells known as glioblastoma stem-like cells (GSCs). Although its origin is still unknown, it is believed that there is a strong association withneural stem cells (NSCs) transformation. Further, it is known that modulation in prion protein (PrPC) expression levels interferes in the biology of GSCs and that the interaction between GBM and NSCs is mostly mediated by extracellular vesicles (EVs), strongly contributing to increased tumor aggressiveness. Given this, GSCs cultivated as neurospheres from PDX cells expressing different levels of PrPC will be initially exposed to EVs from NSCs and then submitted to treatment with temozolomide. Through the evaluation of cell viability, we will know if PDX cells previously treated with EVs from NSCs become more sensitive to chemotherapy. (AU)
