Effect of systemic Bauhinia forficata and biomolecules applied to titanium implant surfaces on periimplant bone dynamics of type 2 diabetic rats: a translational study.

Type 2 diabetes (DM2) affects more than 90% of all patients with comorbidity and requires greater care during titanium implant rehabilitation due to higher rates of failure and postoperative complications. This project to evaluate the effect of systemic and local therapies on the peri-implant repair process, aiming to control negative effects promoted by type 2 diabetes, reducing the inflammatory response as well as other factors that interfere with the formation and mineralization process of the reparational bone tissue. One hundred and seventy-six rats will be used, divided into four groups (n = 44 per group): 1- NG: normoglycemic rats; 2- NGBf: normoglycemic rats treated with Bauhinia forficata; 3- DM2: type 2 diabetic rats; and 4- DM2Bf: type 2 diabetic rats treated with Bauhinia forficata. Each experimental group will be divided into two subgroups according to the surface treatment of the implants (n = 22 per subgroup): Ti - conventional grade IV titanium implants with double acid attack; and vD - grade IV titanium implants functionalized with vitamin D. DM2 will be induced by the cafeteria diet in association with low dose of streptozotocin. NGBf and DM2Bf animals will be treated daily with Bauhinia forficata tea (150 g/L). Two weeks after the beginning of the treatment with tea, the animals will be submitted to implant installation surgery (Ti or vD). The euthanasias will be performed 7, 14 and 28 days after the surgery. The following analyses will be performed: 7 days - immunohistochemistry and maturation of collagen fibers; 14 days - RT-PCR, biomechanics, immunohistochemistry and maturation of collagen fibers; and 28 days - RT-PCR, biomechanics, immunohistochemistry, maturation of collagen fibers, microtomography, confocal microscopy and static histometry. The quantitative data will be submitted to the normality test and this will indicate a more adequate statistical test (parametric vs non-parametric). It will be adopted for all test significance level of 5%.
