Synergic antibiofilm action of the combination of vegetable extracts on Candida species of dental interest

Abstract

Candida spp. they are common microorganisms to be found in the oral cavity, however, they are opportunistic pathogens and the epidemiology of Candida infections has changed with non-albicans species. Phytotherapy has emerged as an alternative to the treatment of oral candidosis and the investigation of the synergism of natural or synthetic substances has shown promising results. The present study aims to evaluate in vitro: a) the antifungal potential of glycolic extracts of rosemary and turmeric isolated on planktonic cultures of Candida albicans, Candida dubliniensis, Candida tropicalis, Candida krusei, and Candida glabrata; b) the possible synergism between the extracts and the antifungal potential that such a combination will produce on planktonic cultures of such Candida species; c) antibiofilm action of combinations that are synergistic on monomicrobial biofilms of Candida spp. with three different contact times: 5 min, 30 min, and 24 h. For the analysis of the isolated antifungal potential of the extracts, the broth microdilution method will be used (standard M27-S4 of the Clinical and Laboratory Standards Institute). For the evaluation of the synergism of the extracts, the "checkerboard" technique will be used and the fractional inhibitory concentration index (ICIF) will be adopted. The concentrations that are synergistic will be analyzed on biofilms formed for 48 h in microplate wells for further analysis of cell viability. The results will be statistically analyzed by ANOVA and Tukey Test (p d 0.05). (AU)
