Analysis of vitamin D levels in mice treated with high potency bisphosphonate and submitted to tooth extraction

Abstract

Containing-nitrogen bisphosphonates are predominantly prescribed to control bone resorption; however, they can also interfere in immune system and metabolism of the individuals that are under chronicle use of these drugs. In these conditions, invasive procedures in the oral cavity increase the risk factors for the development of the osteonecrosis of the jaws (ONJs). Vitamin D influences bone metabolism homeostasis and can help in ONJ prevention since it can act in some related facts to its pathogenesis: angiogenesis, inflammation, and bone cells activity. In this way, this study aimed to analyze vitamin D levels of male 129Sv-EV mice treated with 0.25 mg/Kg of zoledronic acid (ZL) (Group ZL), in order to compare them with animals treated with 0.9% sterile saline solution, both via IP every 72 hours. After 4 weeks from the beginning of the treatment, all the animals will have their upper right incisor extracted, and the treatment will continue until the 21 st day after tooth extraction, when they will be euthanized for blood and maxillae collection. Serum concentration of 25(OH)D 3 will be obtained by quimioluminescence immuno assay. Maxillae with the dental sockets will be demineralized and paraffin embedded, in order to be sliced in coronal sections and stained with HE for histopathological and histomorphometric evaluations, considering blood clot, collagen fibers, fibroblasts, blood vessels, leukocytes, bone matrix, osteoblasts and osteoclasts, empty lacunae, and bone sequestrum. For this, 10 histological fields under 100x magnification will be analyzed for quantification using a grid. Obtained values will be calculated considering area density (%) and treated by statistical tests considering p<0.05. (AU)
