Bone reconstructions with different formulations of Cerabone associated with Raloxifene by sonochemical Route: histological e immunohistochemical characterization in rat calvaria

Abstract

The use of bone substitutes is largely consolidated in bone loss. By functionalization it is possible to promote the improvement of these substitutes, starting from the introduction of bioactive molecules that act directly on the bone cells, triggering favorable responses to the tissue repair process. Good results in this repair have been found with the incorporation of raloxifene into biomaterials, using the sonication process. The objective of this project will be to evaluate, through histomorphometric analysis, immunohistochemistry, and computerized microtomography, a surface alteration, through the homogenization and functionalization promoted in the Cerabone® Biomaterial combined with Raloxifene by means of the chemical sonication method, in calvarias of rats. For this, 96 male rats will have an induced bone defect in the calvaria, where the biomaterial will be used as a graft. They will be randomly divided into 6 groups (n = 10), according to the graft material, Group 01 (Autogenous Bone), Group 02 (Cerabone®), Group 03 (Cerabone® sonicated), Group 04 (Cerabone® and Raloxifene sonicated), Group 05 (Cerabone® and gel vehicle), Group 06 (Cerabone® and Raloxifene gel). After a period of 14 and 28 days after surgery, the animals will be euthanized to remove the specimens to be analyzed. The data will be submitted to the normality curve to determine the appropriate statistical test in the software GraphPad Prism 7.03, with the adoption of the significance level P <0.05. (AU)
