Abstract

Inflammatory bowel disease affects 11 million people over the world. If the quit diagnosis is not obtained and not properly treated, they can lead to the development of colorectal cancer, one of the most recurrent in Brazil. One of the promising natural substances to treat such diseases is 4-methylsculetin (4-ME), from the coumarin class. In this project, it is intended to enhance the anti-inflammatory action of the coumarin 4-ME in the treatment of inflammatory bowel diseases by encapsulating this compound in nanoparticulate lipid systems. Such systems may allow the most effective delivery of the substance in target tissues as well as its controlled release, enabling an increase in the treatment effectiveness. For this, two carrier systems widely used for such purposes will be compared: solid lipid nanoparticles (SLN) and liposomes. Characterization techniques for lipid systems will be used, such as optical spectroscopy, nanoscopy, dynamic light scattering and zeta potential, as well as the study of encapsulation efficiency. In addition, in vitro studies of controlled release and cell biocompatibility are planned in partnership with UNESP of Botucatu city. This work will be complemented using computational methods (artificial intelligence) and mathematical models to determine the mechanism of coumarin release from nanoparticles. This postdoctoral project is linked to the FAPESP Thematic "Towards the convergence of technologies: from sensors and biosensors to the visualization of information and machine learning for data analysis in diagnosis", 2018/22214-6, and will be developed at the Laboratory of Nanostructured Materials for Environmental and Biological Analysis, coordinated by Prof. Dr. Carlos José Leopoldo Constantino at UNESP in Presidente Prudente. In addition, a 12-month BEPE internship will be held at the University of Southern Denmark, Odense, Denmark.
