Effect of the herbal medicine Abuta grandifolia on acute kidney injury induced by ischemia: a preclinical trial

Abstract

Acute kidney injury (AKI) is clinically defined as a 0.3mg/dL or 1.5-fold increase in baseline serum creatinine in 48 hours or a reduction in urinary output to less than 0.5mL/Kg during 6h. Ischemic AKI (iAKI) is predominantly triggered by renal hypoperfusion related to events such as kidney transplantation, unfavorable hemodynamic conditions and even sepsis. Experimental studies demonstrate renoprotective effects of herbal medicines on AKI. In Brazil, given the great Amazon biodiversity, Abuta Grandifolia (Abuta), widely used empirically in the Amazon population for the treatment of kidney diseases, stands out for its anti-inflammatory effects with a possible renoprotective action on AKI. Materials and methods: This is a descriptive, prospective, quantitative and experimental approach with adult male Wistar rats, weighing between 250-300g, randomized into groups: Control (SHAM, with simulation of renal pedicle clamping); Abuta (Abuta grandifolia, orally (po), 5 days; 300mg/kg); Ischemia (clamping of bilateral renal pedicles for 30 minutes, followed by reperfusion) and Ischemia+Abuta (Abuta treatment followed by ischemia). Studies will be carried out to assess renal function through creatinine clearance (Clcr); inulin clearance (Clin) and oxidation, through the measurement of urinary peroxides, TBARS (malondealdehyde) and thiols.Keywords: Acute Kidney Injury.
