Abstract

This project aims to evaluate the effectiveness of the PILP process associated or not with a resin composite containing particles of DCPD (dicalcium phosphate dihydrate, CaHPO4.2H2O) in the functional remineralization of artificially demineralized human dentin (in vitro phase) or after selective removal of the carious tissue (in vivo phase). The composite formulation will be defined in the sub-project described in work plan PD1. This material will be used in the remineralization of dentin blocks (previously immersed for 66 hours in acetate buffer at pH 5.0), associated or not with pre-treatment of the surface with a solution containing mineral precursors in liquid phase (PILP solution). Four experimental conditions will be evaluated in vitro and in vivo, involving two composites (with and without DCPD) and pre-treatment with PILP solution (with or without pre-treatment). In the in vitro phase, after 24 h, 7 days and 14 days of immersion in simulated body fluid (SBF), the dentinal remineralization at the interface with the restorative material will be evaluated through attenuated total reflectance mid-infrared spectroscopy (ATR-FTIR ) through the qualitative analysis of the spectra and calculation of the mineral:organic matrix ratio. Another set of teeth will be used in the evaluation (through nanoindentation) of the in-depth mechanical properties of the initially demineralized dentin, after 14 days in SBF. The nanoindentation test will also be applied to primary teeth subjected to the same treatments in vivo after exfoliation (see main project). Data will be analyzed using ANOVA and Tukey test (alpha: 5%). (AU)
