Acquired pellicle engineering with CaneCPI-5 added to a topically applied gel against dentin erosion: in situ study

Recently, a new cystatin (derived from sugarcane) demonstrated protection against initial enamel erosion (in vitro). In addition, an in situ experiment using the same protein (added in a solution) demonstrated protection against erosion and erosion associated with abrasion for enamel and dentin through the concept of "acquired film engineering". However, to date, there are no results of the incorporation of this protein added in gel on dentin samples. Thus, this project aims to evaluate the protective potential of acquired pellicle modification with CaneCPI-5, using a gel against dentin erosion in situ. 180 specimens of human root dentin (4 x 4mm) will be divided into two types of procedure: Erosion (n = 90) and Erosion+Abrasion (n = 90). For each of them, the study will be conducted in 3 crossed and double-blind phases, according to the gel treatment: experimental gel with 1.0 mg/ml CaneCPI-5 (n = 30); gel containing 12300 ppm F (NaF: positive control) and placebo gel (n = 30). 15 volunteers will use a palatal appliance for 5 days in each phase. 1 additional treatment will be carried out before inserting the device in the oral cavity (on the first day of each phase). In addition, 4 treatments per day will be performed individually, with a micro brush (for one minute). Subsequently, the erosive challenge will be carried out with 0.1% citric acid (4x per day), followed or not by an abrasive challenge (2x per day). The evaluation of wear will be done by contact profilometry. The data will be checked for normality and homogeneity, for the selection of the appropriate statistical test (p<0.05).(AU)
