Abstract

Acquired Immunodeficiency Syndrome (AIDS) is considered a risk factor for osteoporosis. Among the many causes that lead seropositives to a lower bone density, there is the use of antiretroviral drugs, especially Protease Inhibitors (PI), such as Lopinavir/Ritonavir (LPV/RTV). In vitro, it was able to decrease the osteogenic activity of primary bone cells, with significantly lower levels of alkaline phosphatase (ALP) and mineralization, in addition to generating senescence and increasing oxidative stress in osteoblast precursors. Thus, new therapeutic possibilities that can lessen, if not avoid, these damages to bone tissue, are urgent. In this context, the potential benefits of LED (light-emitting diode) therapy emerge. In addition to being non-invasive, it has a low cost and does not depend on the administration of pills for patients who already use large amounts of anti-HIV drugs. In vitro, LED proved to be able to significantly stimulate the viability and proliferation of osteoblasts, ALP indices, the formation of mineralized nodules, and the expression of genes related to cell differentiation. Thus, in order for led therapy to establish itself as a valid treatment option for HIV-positive patients, it is necessary to confirm that the damages of the PI are not amplified and that the benefits remain. Therefore, in this study, we will investigate the consequences of LED irradiation on osteoblasts treated with LPV/RTV. For this, pre-osteoblasts (MC3T3-E1) will be treated with the drug and irradiated or not with LED under different protocols. Cell proliferation and viability (Crystal Violet and MTT), mineralization (Alizarin Red), and ALP will be evaluated. (AU)
