The aim of the present study is to evaluate the cytotoxicity of co-initiators and its influence on release of cytokines from human dental pulp cells (HDPCs). Cells will be isolated from the dental pulp of sound human third molars. The co initiators DMAEMA, EDAB, EHA and BPI will be diluted in dimethylsulfoxide (DMSO) at different concentrations (100¼M; 250¼M; 500¼M; 750¼M, 1mM e 5 mM). In this way, experimental groups and two controls (without treatment and DMSO) will be obtained. HDPCs (10 x 104 cell per well) will be seemed on 96 well, and incubated at 37o C and 5% CO2 for 24h. After this, the cells will be exposed to the different monomers concentrations cited during 24h. After this time, the culture medium will be removed and the cytokine released (IL-1b, IL6, IL-8, IL-10, TNF-±) analyzed by MAGPIX assay, as well as the cell viability by MTT assay. The obtained data will be tabulated and the statistical analysis will be performed.
