Abstract

Renal cancer is among the 10 most incident carcinomas in the world, with about 403,000 new cases diagnosed in 2018. Recent studies have indicated an increase in the incidence of renal cancer in Brazil. Genome wide association studies (GWAS) are applied to identify single nucleotide polymorphisms (SNPs) in chromosomal regions at risk for the development of phenotypes such as cancer. From these studies, 13 different risk regions have been identified for kidney cancer however the biology of these regions is not fully understood. In this project, we will evaluate the role of SNPs rs7132434 and rs10842708, associated with the risk of developing renal cancer, in the regulating the expression of RASSF8, BHLHE41, ITPR2 and SSPN genes and the mechanisms involved in renal cancer biology. To this end, we will perform assays for repression of functional variants associated with the risk of developing renal cancer using CRISPR interference and functional assays from the overexpression of genes potentially regulated by these variants.
