Abstract

Introduction: Studies involving diabetes mellitus (DM) have been one of the main focuses of researchers in recent years, due to its high prevalence and mortality in Brazil and worldwide. Such a chronic condition is characterized by hyperglycemia - the increase in the concentration of glucose in the bloodstream -, causing several comorbidities such as atherosclerosis, retinopathy, neuropathy and nephropathy. The pathophysiology of DM involves several factors, the most important of which are changes in the functioning of enzymes and membrane receptors, activation of inflammatory processes and increased production of reactive oxygen species (ROS). Esculin is a coumarin derivative classified as polyphenol. According to the latest research, this substance is able to act as a potent antioxidant, improving the lipid profile and reducing the synthesis of pro-inflammatory cytokines. Objective: To study the effect of esculin in mesangial cells of mice submitted to high glucose medium. Methods: Immortalized mesangial cells of mice (MiMC) will be cultured in culture plates with DMEM medium and nutrient F12, supplemented with 5% fetal bovine serum. Subsequently, the cells will be allocated into four groups: NG (control, 6.7 mM D-glucose); MA (osmolarity control, 30mM mannitol); HG (high glucose, 30mM D-glucose); HG + ESC, (high glucose treated with esculin hydrate 10, 25, 50, 100 and 200 µg / mL) for 24, 48 and 72 hours. In these groups, we will evaluate the viability, cell proliferation and antioxidant capacity of esculin and its effect on oxidative and nitrosative stress. Statistical analyzes will be performed using the GraphPad Prisma 6.0 software. The results will be presented as mean ± SE, with significance for p <0.05.
