Cancer is considered a worldwide epidemic today. Breast cancer is the most prevalent neoplasm in women and also in dogs. Tumor cell lines from animal models are used in assays, such as the 4T1 murine breast tumor line, for testing with new medications. Recent studies demonstrate benefits of using dichloroacetate (DCA) and omeprazole (OMP) in the treatment of malignant neoplasms. DCA has an important effect on the Warburg effect on the tumor cell while OMP is able to neutralize the pH of the extracellular medium of the tumor microenvironment, inducing anti-proliferative and pro-apoptotic effects. However, there is a lack of data and trials on the application and effectiveness of the effect of these medications. Thus, the objective of this work is to carry out in vitro tests to evaluate the effect of treatment alone and in combination with dichloroacetate and omeprazole in murine mammary tumor cells in the 4T1 strain. For this, four groups will be created: 1) Control; 2) Dichloroacetate, 3) Omeprazole; 4) Dicloacetate + Omeprazole. Dose spectra of medications in cell cultures will be tested and the growth curve will be analyzed by the violet crystal method. Statistical analyzes will be made by comparing the groups' means by ANOVA test. The synergism between DCA and omeprazole will be carried out with the aid of the CompSyn software.
