Orange (Citrus sinensis) is one of the most consumed fruit in the Brazilian diet and rich in phenolic compounds, manily flavonoids. However, the mechanisms responsible for the beneficial health effects resulting from its intake are still scarce due to the study models, which do not allow the broad evaluation of the biochemical response in a complex organism. The metabolomic and proteomic approach is proposed as a tool to broaden the understanding of the complex relationship between nutrition, bioactive compounds and human metabolism. Thus, the aim of this study is identify the metabolic effects of the consumption of fruit polyphenols using metabolomic and proteomic aproaches and also to characterize groups responsive to the intervention. In addition, identify bioactive metabolites responsible for the biological action and the biomarker of their intake. To achieve this purpose, eutrophic and overweight volunteers (n = 85) intake 500 ml of pasteurized orange juice/day, during 60 days. Plasma and urine samples were collected at time zero (before the intervention), 30 day and 60 day. The procedure of this study was approved by the Ethics Committee of the School of Pharmaceutical Sciences-USP (CAAE 14344819.0.0000.0067) and Hospital Universitário (HU-USP) (CAAE 14344819.0.3001.0076). Plasma samples will be deproteinized and analyzed by mass spectrometer (LC-qTOF) for metabolomic analysis. In addition, plasma samples will also be analyzed by LC-qTOF for proteomic analysis. The metabolomic and proteomic data will be analyzed by bioinformatics and both approach will be integrated. In this context, the application of different 'omics' tools allows a better characterization of the individual phenotype and elucidates the role of food bioactive compounds in the health promotion.
