Development of a bioactive titanium surface by alcalinization: effect on adhesion of metalloproteinases inhibitors and metabolism of human osteoblasts

Abstract

The successful installation of osseointegrated implants depends on the obtention of an effective anchorage of these components to the maxillary bone tissue. This osseointegration is related to the adhesion and metabolism of osteoblasts to the implants surface. Studies demonstrated that surface treatment can improve the interaction of this cells to the titanium surface and accelerate tissue repair. This investigation will evaluate the effect of the alkali-treatment of titanium discs with sodium hydroxide (NaOH) for the development of bioactive surfaces. These surfaces will be associated to a metalloproteinase inhibitor aiming the higher adhesion and metabolism of human osteoblasts and faster bone healing. Titanium machined discs will be obtained and standardized. Then, will be selected for each experimental group according to treatments: machined discs, alkali-treated discs and association with naringenin. Osteoblasts will be seeded onto these discs (2 x 104 cells/ disc), previously placed into 24-well plates, using complete DMEM culture medium. After 24 hours, 48 hours and 7 days cell adhesion and proliferation will be evaluated. The effect of surface functionalization will also be determined by synthesis of metalloproteinases and osteoblasts differentiation. Statistical tests will be selected according to data distribution, considering a 5% significance level. (AU)
