Ozone therapy evaluation in the treatment of zoledronate-induced osteonecrosis in senile rats

Abstract

Ozone therapy has been shown to be a promising tool for the prevention of infections and to aid tissue repair, in line with the challenges in the treatment of medication-related osteonecrosis of the jaw (MRONJ). This project aims to analyze the effects of ozone therapy in 55 senile rats (18 month) with 300g to 350g, through drug induced osteonecrosis (Zoledronate 100¼g/kg) after extraction of the first mandibular molar. The animals will be divided into 4 equitable groups (10 rats per group), the first SAL group, will receive saline applications every 3 days for 7 weeks, the SAL + OZ group will receive saline application every 3 days for 7 weeks and treatment with ozone therapy (0.7mg/kg) every 2 days for 28 days, the ZOL group will receive zoledronate (100¼g/kg) every 3 days for 7 weeks and finally the ZOL + OZ group will also receive applications of zoledronate in the same protocol and will be treated with ozone therapy (0.7mg/kg) every 2 days for 28 days. All rats will receive antibiotic therapy, will be submitted to extraction of the mandibular molar in the third week of experiment and the euthanasia will be at the end of the seventh week of experiment. The study will characterize the architecture of the bone tissue at senility (day 0 of the experiment - MCT0) and after the zoledronate application course (MCT1), compared to the group that will not receive zoledronate (MCT2), in those 15 rats, will be distributed in these three groups (MCT0, MCT1, MCT2), each of them with vive rats destined for the analysis in computerized microtomography (MicroCT). The jaws will be resected, reduced and prepared for micro-CT, histological and immunohistochemical analyzes (TNF-±, IL-1², VEGF, OCN and TRAP). The data will be submitted to the normality curve to determine the most adequate test (P <0.05). (AU)
