Abstract

Dental Caries is a multifactorial disease that leads to the destruction of dental tissue. The difficulty of eradicating this condition demands new approaches to its control. Therefore, the objective is to prepare formulations with crude extracts and fractions (combined or not with fluoride) of Casearia sylvestris (Biomes Atlantic Forest, Cerrado, Ecotone-Cerrado/Caatinga and varieties sylvestris and lingua) and to test these formulations for the control of cariogenic biofilms. Four experimental steps will be performed: (1) collection, preparation, fractionation, and chromatographic analysis (via UHPCL-DAD-HRMS and RMN) of extracts and fractions. This data will be used to prepare formulations (with and without fluoride) for the next steps; (2) analysis of the effect of Streptococcus mutans biofilm formulations formed on salivary film hydroxyapatite discs by assessment of viable microbial population, biomass, and extracellular matrix components (water-soluble and -insoluble exopolysaccharides or EPS, lipoteichoic acid or LTA, and extracellular DNA or eDNA), and 3D architecture. The data from this step will show the best formulation (or formulations) to perform step 3; (3) evaluation of formulations on oral microbiota homeostasis (sequencing) and extracellular matrix quality (EPS, LTA, and eDNA) of microcosm biofilm, and prevention of enamel demineralization (microhardness); (4) for the most active formulations in the microcosm model, the cytotoxicity will be evaluated using EpiOral TM oral tissue culture (MTT assay). For data interpretation, statistical tests will be applied according to distribution adopting a 5% significance level. (AU)
