Liquid Chromatography (LC) is one of the most versatile separation techniques and is employed in a number of relevant areas of scientific knowledge. Its miniaturization, understood here as the reduction of the internal diameter of the columns, followed the introduction of capillary columns in gas chromatography by Golay. Therefore, the current interest is focused on the development of new LC columns, aiming at increasing the efficiency of the technique and implementing the many benefits inherent to it. These mainly include system miniaturization and its implications such as solvent and sample economy and coupling with detection systems commonly used in gas chromatography such as mass spectrometry. Among the columns employed in LC, we highlight the open tubular capillary columns of the type PLOT (Porous Layer Open Tubular) and WCOT (Wall Coated Open Tubular), with few studies on the evaluation and use of new materials and techniques in its preparation. In this context, this research project aims to study new materials for both types of columns, together with an optimization of the column preparation system to be operated in line with miniaturized systems (¼-LC and nano-LC) coupled to sequential mass spectrometry (MS/MS).
