Chronic stress and the oxidative stress levels in the oral mucosa and oral squamous cell carcinomas

Abstract

Exposure to potentially stressful events during childhood may promote biological and psychological changes that persist even in adulthood. A recent study by our team revealed that chronic stress-induced in the early phase of life (ELS) can increase oral cancer occurrence and progression in rats submitted to oral carcinogenesis. However, the mechanisms associated with ELS-induced oral carcinogenesis are poorly understood. Therefore, the aim of the present study will be to evaluate the impact of ELS on oxidative stress in normal lingual tissue and chemically induced oral squamous cell carcinoma (OSCC). We will use male Wistar rats divided into 4 groups: 1) Control; 2) rats submitted to ELS alone; 3) rats submitted to oral carcinogenesis alone, and 4) rats submitted to EPV and oral carcinogenesis. ELS will be induced in groups 2 and 4 by separating the litters from their mothers daily for 3h (from day 1 to 21 postnatal). On the 90 days of age, rats from groups 3 and 4 will be treated with carcinogen 4NQO diluted in drinking water for 120 days to induce oral carcinogenesis. Rats from groups 1 and 2 will have ad libitum access to drinking water. After the experimental period, all animals will be euthanized and tongue tissues from all animals will be collected for histopathological analysis and for biochemical analysis. Oxidative stress levels in the normal lingual tissues (groups 1 and 2) and tumor tissues (groups 3 and 4) will be evaluated by enzyme-linked immunosorbent assay measuring radical superoxide (O2-), hydrogen peroxide (H2O2) and hydroxyl radical (OH-) and total antioxidant capacity (TAC). Specific statistical tests will be used to assess whether ELS alters oxidative stress levels of the normal tongue and OSCC. With this study, we hope to understand new parameters regarding the association between ELS and carcinogenesis.
