Morphological and biomechanical characterization of reparative bone tissue in rats with metabolic syndrome and osteoporosis treated with systemic sodium Risedronate

Abstract

Osteoporosis is a metabolic disease that mainly affects postmenopausal women, with the main cause of estrogen deficiency. Another condition that shares the same causes is metabolic syndrome, characterized by abdominal obesity, dyslipidemia, hyperglycemia, and hypertension. These disorders can lead to decreased bone density and mass, impairing the quality of bone tissue as well as its repair process. To characterize, from a morphological and biomechanical point of view, bone tissue in ovariectomized rats with metabolic syndrome treated with risedronate, a drug used in anti-osteoporosis therapy. 48 rats will be divided into six experimental groups: SHAM: dummy surgery, SHAM SM: dummy surgery with metabolic syndrome, OVX: ovariectomy surgery without drug treatment, OVX SM: ovariectomy with metabolic syndrome, OVX / RIS: ovariectomy associated with treatment with risedronate and OVX SM RIS: ovariectomy surgery with metabolic syndrome associated with risedronate treatment. Thirty days after the osteoporosis induction, the rats will be submitted to drug treatment with risedronate (0.7 Mg / kg / day, weekly). After 30 days, the animals will receive an implant in the tibial metaphysis for biomechanical evaluation of periimplant bone repair. Euthanasia will occur 28 days after implant installation surgery. After the animals are euthanized, the pair of right and left femurs and tibias containing the osseointegrated implants will be removed and submitted to computed microtomography, biomechanics and femur compression analysis. Quantitative data will be submitted to the normality curve for the definition of a parametric or nonparametric test, considering the significance level of 5%. (AU)
