Abstract

Metallic compounds, such as palladium, have attracted great interest from researchers for exhibiting significant anti-cancer activity accompanied by improved bioavailability, efficacy and toxicity characteristics that limit the use of platinum analog compounds in antineoplastic chemotherapy. Despite this, new palladium complexes are being developed, so it is necessary to evaluate the cytotoxic mechanism of these compounds in tumor cells using techniques that better mimic the environment of tumors in vivo such as multicellular spheroids. The present project will seek to evaluate the cytotoxic response of palladium (II) AJ-5 and BTC2 complexes on T47-D breast cancer cells growing as spheroids, focusing on their effects on cell migration and on apoptosis and autophagy pathways. The research will be carried out following monolayer and spheroid cell culture steps, determination of cytotoxicity of Pd (II) compounds, analysis of viability and cell cycle, protein expression evaluation and confocal microscopy imaging.
