Calorie restriction (CR) is a nutritional intervention that has been shown to extend lifespan in a variety species and also to afford metabolic benefits. The mechanisms involved in these CR benefits are not fully elucidated. Data from our group demonstrated that Saccharomyces cerevisiae cells cultured under calorie restricted conditions apart from having a life span extension, showed increased proteasome activity. This increase is not due to increased proteasome amount, suggesting that other mechanisms are involved in calorie restriction-induced increase in proteasome activity. The present BEPE project aims to characterize the effect of CR on proteasome spatial conformation, as allosteric regulation was shown to be a common control mechanism of proteasome activity. In order to perform this structural characterization, we are going to use cryo-EM. (AU)
