Secondary metabolites derived from bacteria, fungi and plants provide a diversity of bioactive chemical compounds that has countless application in the industry segment. Yet, high costs and time-consuming prospection of new molecules employing outdated methodologies discourage the search for these metabolites. Recently, the huge progress in molecular biology methodologies and the breakthrough and low-cost in total genome sequencing have made natural products, manly those produced by actinomycetes that has a vast secondary metabolite, to return to be debated and exploited by techniques knows as genome mining. The genome mining of genes clusters became a powerful tool in the discovery of new chemical entities. Genes exploitation are simplified by computational tools such as BLASTp (NCBI), HMM, Pfam, AntiSmash among others. Theses bioinformatics tools, through complex algorithms, aid in the mining of potential genes clusters that encode lipids, terpenes, siderophore, non-ribosomal peptides, polyketides and other chemical that has considerable economic interest, which are not always observed in the culture medium under laboratory conditions. Amidst secondary metabolites, terpenes are the largest class of natural products with wide structural and chemical diversity. These natural products display diverse ecological functions, as well as commercial applications with high value-added. Thousands of terpenes are present in terrestrial and marine plants, as well as fungi. Nonetheless, it has recently been found that a variety of bacteria, mainly gram-positive Streptomyces and other actinobacteria, use their biogenetic machinery to design this class of secondary metabolites, becoming a valuable source in the discovery of new natural products. This project aims to study the functionality of terpenes biosynthetic genes clusters encoded in the endophytic strain Streptomyces sp. CBMAI 2042, enlightening steps involved in the biosynthesis of these metabolites through the characterization of the enzymes terpenes synthases and the respective natural products obtained by enzymatic reaction assays.
