PHBV nanofibers containing sol-gel derived bioactive borate glasses doped with therapeutic ions for tissue regeneration

Abstract

As a result of skin lesion generated by the most diverse causes, tissue engineering has developed synthetic grafts capable of promoting the cellular regeneration acceleration, lower risk of infection and greater degree of comfort to patients in recovery. Grafts composed of poly (3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) nanofibers have several characteristics necessary for application, but the lack of bioactivity becomes a limiting factor. The incorporation of bioactive glasses into the nanofibers is capable of promoting the bioactivity required for the graft since it forms a layer of carbonated hydroxyapatite [CHA, C3Ca10H2O23P3] (similar to biological apatite) on its surface, which chemically bonds to both soft and hard tissues. Since bioactive borate glasses exhibit higher conversion rates in HAC than silica based, formulations such as 45B5 (46.1% B2O3, 26.9% CaO, 24.4% Na2O and 2.6% P2O5, % mol) become promising for incorporation into PHBV nanofibers. The synthesis of bioactive borate glasses via sol-gel allows the glass structure modification with therapeutic ions with greater ease than the fusion-cooling method, such as ions capable of promoting angiogenesis (induce vascularization) (Co2+) and the antimicrobial activity (Zn2+), or even both of these therapeutic functions (Cu2+). In this context, the this work main objective is the production of PHBV nanofibers coated with bioactive borate glasses nanoparticles modified with Co2+, Cu2+ and Zn2+ by sol-gel method, by means of electrospinning and electrospraying techniques, allowing the production of nanofibers coated with nanoparticles, besides the obtained products characterization.
