Analysis of blood markers in patients with Osteosarcoma, Soft Tissue Sarcomas and Desmoid Tumor

Abstract

Circulating Tumor Cells (CTCs) are rare cells that detach from some tumor site, invade blood vessels, being able to achieve new sites to form single or multiple micrometastasis. Desmoid tumor, also known as aggressive fibromatosis, is a rare, locally invasive, and non-metastasizing type of cancer. Soft tissue and bone sarcomas include a large group of several heterogeneous subtypes of mesenchymal origin, they are rare and aggressive tumors. Searching and finding CTC in these groups, the expression of proteins in these cells and mutation analysis may contribute to a personalized treatment for these patients. Objective: quantify CTCs, specific protein expression and mutations by ddPCR, and establish primary culture of these cells. Methods: the blood (approximately 12mL) will be prospectively collected from patients with desmoid tumor and sarcoma. The samples will be processed and filtered on ISET (Isolation by Size of Epithelial Tumor Cells, Rarecells, France) technology for the isolation of CTCs. For quantification and characterization will be performed immunocytochemistry (ICC) and counterstained CTCs with hematoxylin-eosin, for mutation analysis will be performed ddPCR. Expected Results: We hope that this study can show if the CTCs can be used as "biopsies net" both in patients with desmoid tumor and sarcomas. (AU)
